Ribosomal Synthesis of Thioether-Bridged Bicyclic Peptides.
Many biologically active peptides found in nature exhibit a bicyclic structure wherein a head-to-tail cyclic backbone is further constrained by an intramolecular linkage connecting two side chains of the peptide. Accordingly, methods to access macrocyclic peptides sharing this overall topology could be of significant value toward the discovery of new functional entities and bioactive compounds....
متن کاملTranslocation of a thioether-bridged azurin peptide fragment via the sec pathway in Lactococcus lactis.
This study demonstrates for the first time that a thioether-containing peptide, an azurin fragment, can be translocated via the Sec pathway. This methyl-lanthionine was introduced by the nisin modification enzymes. The Sec pathway can therefore be a successful alternative for those cyclized peptides that are inefficiently transported via NisT.
متن کاملBioinspired Strategy for the Ribosomal Synthesis of Thioether-Bridged Macrocyclic Peptides in Bacteria
Inspired by the biosynthetic logic of lanthipeptide natural products, a new methodology was developed to direct the ribosomal synthesis of macrocyclic peptides constrained by an intramolecular thioether bond. As a first step, a robust and versatile strategy was implemented to enable the cyclization of ribosomally derived peptide sequences via a chemoselective reaction between a genetically enco...
متن کاملStructural characterization and proton reduction electrocatalysis of thiolate-bridged bimetallic (CoCo and CoFe) complexes.
Using an assembly method, dinuclear CoCo and CoFe complexes supported by a bdt ligand, [Cp*Co(μ-η2:η2-bdt)(μ-I)CoCp*][PF6] (1[PF6], Cp* = η5-C5Me5, bdt = benzene-1,2-dithiolate), and [Cp*Co(μ-η2:η4-bdt)FeCp'][PF6] (3[PF6], Cp' = η5-C5Me4H) were synthesized in high yields. Upon chemical reduction with CoCp2, complexes 1[PF6] and 3[PF6] were converted to [Cp*Co(μ-η2:η2-bdt)CoCp*] (2) and [Cp*Co(μ...
